Has anyone taken Vraylar? If so what kind of results did it give you?

I don't know. I have chickened out after 2 days. These meds scare me. Experience has shown that it can really go sideways. The reason I don't generally recommend meds to each depressed individual is that you don't know in advance what will happen. If you knew, Id recommend them almost universally.

No, but I'm taking Reagila, which has the same active principle, cariprazine. I'm taking 3 mg.

Pretty much nothing at all. At first, maybe it was a bit activating, but I may be mistaken (could be placebo). If anything, 1.5 mg was better.

I see it all the time on commericals, if you live in the US. It's for bipolar I

I tried Vraylar a couple of years ago. I had been waiting for Vraylar to come out for 1-2 years, and I was optimistic. Unfortunately, I felt significantly more depressed almost immediately. That was a bad sign for me. I elected to discontinue Vraylar at that point - mainly because of how extraordinarily long its half-life is. If I continued to take it, and I felt worse and worse, it might have taken weeks to get out of system. I was unwilling to take that risk.

Some people have done well on Vraylar. I'm guessing you have already tried Abilify? I was interested in Vraylar because it the ratio of D3/D2 binding is significantly higher than Abilify.

That is its unique property, and the main one why I was interested in trying it too. That said, blocking dopamine D3 is not necessarily a good idea -- possibly even the opposite. Pramipexole, a D3 *agonist* was the only drug that ameliorated my anhedonia.

When my psychiatrist told me about v , it sounded like a chemical straight jacket. Blocks d2 with less agonist activity than abilify.

Rexulti, on paper, looks better for maintenance...to me, at least.

I never felt like Neuroleptics were a "chemical straight jacket". I still have feelings... They just don't overwhelm me anymore.

Abilify and Vraylar bind to D3/D2 receptors, but they are not full antagonists. They are partial agonists. When Abilify first appeared, Stephen Stahl characterized it a dopamine system stabilizer.

1. High dopamine in the synaptic cleft -> receptors are antagonized.

2. Low dopamine in the synaptic cleft -> receptors are stimulated.

It sounds weird, but I imagine there is some kind of conformational change in the receptor at high versus low dopamine concentrations. The postsynaptic dopamine receptor actually has two distinct states that have different binding affinities. This might explain the dual action of partial agonists.

That's interesting. Maybe that's why it began to make me feel very much worse.
