Peaking would mean the time it takes for a drug to reach its highest concentration in the blood stream[plasma levels] after which the level of the drug will begin to drop. Therefore a drug with a short half-life would peak quite rapidly. Onset of action means the time it takes for a drug to take effect. There is also steady state concentrations. This means the time it takes for a steady level of a drug to be in the body. In some cases this can take weeks. Hope this helps.
