Predicting the efficacy of an antidepressant must take into consideration more than just its action at membrane bound transporters as observed in vitro in synaptosomes. As an example of this is that the distribution of these drugs in different regions of the brain must also be considered. Atomoxetine selectively inhibits the reuptake of norepinephrine as does desipramine. However, atomoxetine seems to accumulate in prefrontal cortex whereas desipramine does so more globally. Desipramine is a very effective antidepressant while atomoxetine is not.
