Hi, Uncouth.

> Scott, why would you go off of Parnate to try Brintellix? Have you heard differentially of THAT much benefit Brintellix can provide?

I am stuck. I guess I want what I once had. I might change my mind along the way if I become desperately depressed again. I am down from 120 mg/day to 100 mg/day to 80 mg/day. I'll probably stay there for a few days.

I have been impressed by the few success stories I have seen here regarding Brintellix. Apparently, when it works, it works well. My doctor has had a few of his TRD patients respond to it. My short term plan is to discontinue Parnate and try Brintellix. I should probably have a colonoscopy while I'm off of a MAOI.

When it worked, Nardil made me feel better than Parnate does. I don't know. Maybe high dosages of another hydrazine would give me more motivation and resolve anhedonia, two symptoms that Parnate doesn't help with.

> I'm assuming it's the 5ht7 antagonism..,but your on abilify already right?

Yes. :-) You really know your stuff. Brintellix is also a full agonist of 5-HT1a receptors rather than a partial agonist as is Abilify.

5-HT reuptake inhibitor
5-HT1a full agonist
5-HT1d partial agonist
5-HT3 antagonist
5-HT7 antagonist
5-HT1b antagonst

I will also consider switching from Abilify to Vraylar. I am guessing that it should increase motivation and mental energy more than Abilify due to its higher selectivity for dopamine D3 receptors as a partial agonist.

> If more of that is what your seeking have you tried adding low dose amisulpride?

That's an interesting idea. Thanks.

> How high have you pushed the Parnate?

150 mg/day. It didn't help any more than 120 mg/day of Parnate. The thing is, one can experience a brief improvement in vegetative symptoms whenever the dosage is increased. Perhaps this is due to its stimulant-like properties. This improvement can deceive one into dosage escalation. I will let you know how I feel as I reduce the dosage. There will probably be a lag time between dosage reduction and relapse, as it may take a few days for new MAO to be synthesized.

> I am just escalating (again) after having to stop 120mg Parnate over two weeks two months ago. I will continue to push it as high as I need to this time around as I finally detected partial effectiveness when I hit 120mg before surgery.

> Oh and I tried Marplan earlier this year in between Nardil and Parnate....due to Nardil weight gain. I think I went as high as 80 or 90, and it didn't feel as effective as Nardil but I wish I had pushed the dose.

Did Marplan help at all? If so, which symptoms did it improve?

What is the longest time you remained at one dosage of Parnate? Adding lithium 300 mg/day or Lamictal might help.

I will play with resveratrol and N-acetylcysteine (NAC) at some point.
