Why these meds don't work

Posted by linkadge on August 5, 2004, at 17:34:36

reputake inhibitors are extremely blunt. SSRI's increase serotonin at all receptors.

There is perhaps, only one or two receptors that benefit from serotonin agonism. When other receptors recieve serotonin other devastating side effects can occur.

Agonism of the 5ht-2a receptor makes people very nervous and makes sleep broken and full of nightmares it is proposed that agonism at the 2a receptor can make people suicidal too, agonism of the 5-ht2c receptor causes sexual dysfunction and frontal cortex hypofunction/cognative problems. Agonism at the 5-ht3 receptor causes GI problems and and hippocampal kindling/panicky problems. As well it can make people antisocial.

Agonism at the 5-ht1b receptor causes cognative impariment.

I just cant do anything. If I take them I feel like crap, if I don't take them I feel like crap.

Buspar is on-point, it is a partial agonist at the 5-HT 1A receptor, which modulates serotonin release. Supposedly, if levels are too low, it will raise them, or if too high, it will lower them. I have read that long-term use (>8 weeks) desensitizes this receptor, leading to stronger serotonin release.

The strange thing about Buspar is that it was marketed as an anxiolytic, not as an antidepressant. I have sometimes wondered that if Buspar had received the same hype that the SSRI's did, it would be considered as strong an AD as Lexapro, Paxil etc. Its close relative gepirone was due for approval a few weeks ago, but failed to get it from the FDA. It was to be marketed as an antidepressant.

I have heard people getting an antidepressant effect when it was taken in higher doses.

I sympathize with this sentiment. Especially now that Prozac isn't doing what it used to for me. It used to help me make connections between ideas, be more playful, hate myself less...

I didn't know that 5HT agonism was implicated in cognitive impairment. That really stinks, 'cause that sort of thing is the primary symptom of my depression.

I wish I knew whether my problems were caused by swallowing these compounds for so long or by quitting too many times, causing some kind of "kindling," similar to what happens in epilepsy.

Not that knowing would change much at this point. Like you I'm dependent on meds.
